Basically you will look in the vehicle owners manual for the towing capacities of that vehicle. Also there should be listed a gross combined weight rating. This is the maximum that the vehicle and trailer can weigh together and includes cargo, passengers, fuel and anything else in the vehicle and trailer.
Stay within the towing capacity and stay within the gross combined weight rating and you will be fine.

I did contact the manufacturer on your behalf and clearance as far as distance from the bottom of the bumper to the top of the receiver is less than 1/2" for both, so no difference between them. A smaller 1-1/4-inch Class II hitch on your 2008 Toyota Sienna will have a slightly greater height above ground than a 2-inch Class III type.
