I do have a solution for you, however the Curt T-Connector Vehicle Wiring Harness # 55540 referenced in your question is designed to fit the 2003 to present Chevy Express Vans. This wiring harness will not work on your 2001 model.
For your 2001 Express Van 3500, I instead recommend the Curt T-Connector Vehicle Wiring Harness # C55377. This is confirmed to fit your vehicle and will install by connecting directly to built-in ports located behind the trim panels in the cargo area on the driver and passenger side. This will provide you with a 4-Way connector to connect with and power your trailer's signal lights.
