The measurement you will need to start the installation of the base rails in your truck bed is from the rear edge of the truck bed to the rear edge of the base rail closest to the tailgate. That measurement for a 2000 Ford F-350 Super Duty Long Bed is 32-1/4 inches. This is where the rear base rail of the part # RP50082-58,will be installed.
