Can 2 Pole Wring Connector Like PK12200 Be Used to Connect Trailer Side Marker Lights

Can I use a 2 pole connector to attach the side markers? One wire to main line and the other to side markers.

You could indeed use a 2-pole connector like part # PK12200 to connect side marker lamps such as # MCL76RB. One lead would tie into the running lamp circuit, the other would ground to the trailer frame. Using a 2 pole connector would provide a quick disconnect that would allow you to quickly replaced a damaged lamp without needing to cut and splice anything.
Using dielectric grease like # 11755 on your electrical connections is recommended to exclude moisture from the connection and to prevent corrosion.
