Technically two of the Coil SumoSprings Custom Helper Springs # CSS-1195R that you referenced would fit in a single coil but there are a couple reasons this won't be a great idea. First one is that if you were in a situation where you were still sagging significantly after installing one set you are probably overloading the vehicle. And the second reason is that the ride quality would be significantly harsher if you did this.

So if you are sure you are under you towing capacity a single set of these would be all that you need.

There is no difference between the two. Part #CSS-1195 can be used as either front or rear springs depending on the vehicle, and for your 2006 Toyota Sienna they are for the rear springs and as such are classified as #CSS-1195R.
