Question:
does it fit 2001 E350 Van?

Brake controllers like the # C49UR Curt Assure aren't a vehicle-specific item. You can install a brake controller on just about any vehicle.
What the installation will involve will depend on whether your E350 is equipped with a trailer connector and what type it is.
If you have the factory 7-way trailer connector, it'll be a plug-and-play installation using the # C51322 brake controller harness. The harness plugs in to the connector under the dash near the e-brake pedal (see provided photo).
If you have a factory 4 pole trailer connector, you'll still need the # C51322 harness, but you'll need to use a # 37185 adapter to convert the existing connector to a 7-way. You'll also need to sever the blue wire on the
brake controller harness, and graft on a length of # 10-1-1 10 gauge wire long enough to run through the firewall and under the vehicle to connect with the blue wire on the adapter.
If you have no trailer connector, the connector under the dash won't be present, and you'll need to use our # ETBC7 brake controller install kit along with a 4 pole connector like # 118344 to access the taillight circuits without having to splice anything.
I'll link to a help article that explains all this.
