Will this hitch work with an existing bed rail tool box installed in the front of my truck bed? I have a 2017 Ford F-250 with a 6-3/4foot bed.

Yes as long as you have 3" from the back of your tool box to the front edge of your prep package hole the Curt Q20 5th Wheel Trailer Hitch w/ Slider for Ford Towing Prep Package part # C16530-16020 will workout well.
