The brake signal wire on a vehicle that you will connect to the RED input wire on a brake controller like the Prodigy P2# 90885 is the wire that carries a signal only when the pedal is pressed. Some wires at the brake light switch may carry a signal at other times. You want to find the wire on your 2015 Toyota Highlander that has a signal only when the pedal is pressed. Auto makers may change the color of a given wire between production runs, so it is best to use a circuit tester like PTW2993 to confirm which wire carries the brake light switch signal. Often the wire you're looking for is the larger of two green wires so start your testing there.
The other wires on the controller connect as follows: the Black wire takes in +12V power from the battery (through a circuit breaker); the white wire grounds to the battery negative terminal; and the blue output wire is routed to the brake controller circuit on the 7-way. The linked photo shows you the common circuit configuration for a RV-style 7-Way; the trailer brake circuit is carried by the contact at the 5-o-clock position as you view the trailer connector.
A video is linked that will be helpful in guiding your installation.
