It shouldn't be a problem to hook up a 100 lb cylinder to your trailer while it is stored for the winter. As long as you have the correct fittings, you'd just need to disconnect your dual tanks from the trailer's propane line and close them, and make sure your 100 lb cylinder is connected to your trailer's propane line with a dual-stage regulator and a POL hose.

It's likely that the pigtails you already have with your smaller dual cylinders won't work with the 100 lb cylinder, so you need something along the lines of the JR Products # 37207-30675 which has a POL fitting for your larger cylinder, and an inverted 1/4 male flare on the other end to fit into a dual stage regulator such as the Camco # CAM59323 . You'll also need an adapter for that regulator's NPT fitting on the inlet # CAM59953 . Then you'd hook up the regulator to your camper's propane supply using your existing supply hose.
