Yes, the Thule UpRide Roof Bike Rack for Fat Bikes # TH599000-FB will work with tires that are up to 5 inches wide, and is a wheel-mounted bike rack so you do not have to take off your tire when you load the bike on your roof.
The UpRide will hold your bike very securely with a wheel cage and hook for the front wheel and ratchet strap for the rear wheel. You can mount it on your crossbars with its clamp system, or if your roof rack has a channel system (a T-slot), it can also mount that way.
The Thule UpRide Roof Bike Rack for Fat Bikes # TH599000-FB has a hefty 44 lb weight capacity and is extremely durable - so much so that the rack comes with a lifetime warranty, so you'll never need another roof-mounted rack.
I do recommend adding the Thule One-Key System Lock Cylinders # TH450200 to protect your investments; these cylinders will secure your bike to the rack and the rack to your crossbars.
