Does 12 Volt Accessory Circuit of 7-Way Have to Activate for Brake Controller

Is the 12 volt hot lead on a 7 way connector required to power electric trailer brakes?

It is not needed to wire up the 12 volt power circuit of a 7-way to use trailer brakes. Those two circuits are totally separate. If you don't have a need for a 12 volt accessory circuit you would not need to activate it and the controller will work just fine.
