Hello, I was wondering if part #40-3605 requires the removal of the factory-installed front skid plates on the 2016 4Runner?

You would not have to remove the skid plate. I checked with Westin and they do not have any notes about removing the skid plate of a 2016 Toyota 4Runner nor is there any mention of it in the install instructions. I attached an install photo from the instructions that shows that the Westin Sportsman Grille Guard # 40-3605 that you referenced actually attaches to the vehicle through the bumper and not from underneath the front end where the skid plate would be.

The arrow points to the mounting hardware.
