Will the drivetrain management computer of a 2022 Toyota 4Runner need to be re-flashed once a brake controller is installed?

Installing a brake controller won't have any effect on the computer of your 2022 Toyota 4Runner, so you can install any brake controller you wish.
Although the # C51180 Curt Echo wireless brake controller would be a good choice, I'd recommend the under-dash version, part # C27JR instead. The controller would install to the vehicle using the Curt # C51362 vehicle specific wiring harness that would plug into the tow package port under the dash located under the dash just above the kick panel as shown in the linked install video. The Echo under-dash model communicates wirelessly with your smart phone, but will be located inside the vehicle, so there's no danger of it being stolen unlike the # C51180 Echo which plugs into the 7-way trailer connector.
