There is a Class III trailer hitch currently available for the 2021 Toyota Sienna, which is part # C22UR. It doesn't require any drilling and bolts right on to the frame. There is a small amount of underbody panel trimming that needs to be done, but the installation instructions I attached outline the process very clearly.

This hitch has a 2 inch receiver, a tongue weight of 350 lbs and a towing capacity of 3,500 lbs. You will want to keep that in mind because that is at the top of the max towing capacity for a fully equipped Sienna. Be sure to check your owners manual for your Sienna's specific towing and tongue weight capacity before you tow or carry any loads. The lowest capacity between the hitch and vehicle will be your limit.
