Ladder Rack and Tonneau Cover for 2020 Jeep Gladiator

Would this work with a bed rack that mounts under the rails as well?

If you have a ladder rack on your 2020 Jeep Gladiator that utilizes the inside of the bed rails, the TruXedo Lo Pro Soft Roll-Up Tonneau Cover # TX523201 that you referenced won't be compatible. This is really going to be the case with any tonneau cover, as they are going to require this same mounting space for installation.
At this time your only option for a tonneau cover and ladder rack combination will be with an inside-the-rail tonneau like the already-mentioned TruXedo Lo Pro # TX523201 (which is a really nice choice) and the Putco Venture Tec Overland Truck Bed Rack # P184500. This is the only ladder rack for your Jeep Gladiator that sits right on top of the bed rails, leaving the insides of the rails free for the tonneau cover hardware.
This is actually a really nice rack that's specifically designed, both in terms of fit and style, to match your Jeep Gladiator. It features built-in channels on the crossbars that will work with just about any roof bar accessory, integrated tie-down points to let you easily secure your gear, and has a 500 pound weight capacity. A tent-mounting kit is also included hat works with most rooftop tents.
