Recommended Plug and Play 7-Way Connector for 2000 Ford F-250

Will the factory wire harness plug directly into 7 and 4 pin connector HM40975-11998 on a 2000 Ford F250 SD?

The Hopkins 7-Pole and 4-Pole Trailer Connector Sockets # HM40975-11998 referenced in your question will work on your 2000 Ford F-250, however you will need to hardwire the pigtail end to the factory harness on the truck. This will not plug directly into a built-in port on the truck.
An easier solution if your F-250 has the factory 4-Way is to use the Tow Ready Ford OEM Tow Package Wiring Harness # 118243. This will connect directly to the standard OEM connector on the truck and provide a 7-Way connector for your trailer. If you still need to use the 4-Way occasionaly, you can add the Pollak 7 Pole to 4 Way Connector Adapter # PK12716, which will simply plug right into a 7-Way and give you a 4-Way on the other end when needed. This is the setup I recommend for a plug and play option.
