Should Trailer Mounted Batteries Be Grounded to Trailer Frame  

Question:

Looking to run 2, 6 volts batteries in series for house batteries, and I have a question about grounding, should I run a ground cable directly for the 12v neg of the batteries to the frame of the camper? I’m asking this because there appears to have never been a ground to frame cable before on the old 12v battery? And if I’m thinking about installing a battery cut off switch, does that get installed on the pos or neg side?

The batteries should indeed be grounded to the trailer frame. A battery disconnect switch like # 277-000096 would install inline on the positive circuit of the battery as shown in the video I've linked for you.
