It certainly will. According to Curt Manufacturing, the # C13200 Class III trailer hitch is indeed a fit for all models of the 2016 Toyota Highlander.
If you'll be using the hitch for towing purposes, you might also need the following accessories:
Trailer wiring harness, part # 118449
Ball mount, part # C45030
1-7/8 inch trailer ball, part # 19260
2 inch trailer ball, part # 63845
