There is one hatch mounted two bike rack available that is compatible with the 2010 Highlander. The Hollywood Racks Over-The-Top bike rack, part # HRF2 will fit over the spoiler on top of the hatch, so the bike rack mounting straps don't directly contact the spoiler which prevents any damage. It is not recommended that the hatch be operated with the rack in place, as the extra weight of the rack might cause the hatch to close unexpectedly, causing injury to anybody who might be in the way.
If you'll be transporting women's bikes or bikes with sharply-angled top tubes, you might also consider a frame adapter bar like part # 64005. The bar spans the distance between the handlebar gooseneck and the seat post, which provides a secondary attachment point for the bike rack. This allows the bike to be carried level, which maximizes ground clearance.
I've linked you to a product demonstration video for the bike rack, as well as for the frame adapter bar.
