Does the positive post hook to charger and ground to motor on 36 volt golf cart

The CTEK MULTI US 3300 12V Battery Charger # CTEK56353 is designed for 12V batteries and will not work well to charge the 36V battery on your golf cart.
For a 36V battery charger we do have the NOCO Genius UltraSafe Industrial Battery Charger # 329-GX3626. This is an excellent battery charger that features 6 different modes that you can switch to with just the push of a button. You will connect the positive (red) battery clamp or eyelet to the positive battery terminal. The negative (black) battery clamp or eyelet will go to the negative battery terminal or vehicle chassis.
