The hardest part of replacing a roof vent is definitely removing the old sealant. You'll want to use some composite tools, like part # PT73HJ, to carefully pry up the sealant without ripping up the roof membrane. You won't get all of it off but will be able to get a majority. Once you're done prying up the sealant you can wipe down the area with some rubbing alcohol to clean and prep it for new sealant.

The procedure is actually simpler than you think.
Take one of the pull wires and thread it onto the carriage bolt. then, feed the end of the wire through the spacer. You'll then push the bolt, then the block up into the hole in the underside of the frame member. Once both the bolt and block are up into the frame member, gently (Gently!) pull the wire so that the bolt threads into the block and the threaded end of the bolt comes back out the frame hole. I checked with Kat's and the Heater Part # KH11812 that you referenced is designed to fit in the cartridge that's in the RR or rear right position. This is the cartridge that is right around the back of the block on the passenger side of the vehicle.
So if you were facing the engine with the hood up it would be the back corner of the engine on the passenger side.
