It sure will!
The etrailer Trailer Hitch Receiver # e98851 you were looking at for your 2004 Acura MDX is designed to be compatible with a full sized spare tire.
If you are planning on towing with your MDX you will also need a vehicle wiring harness # 118336, and a ball mount, hitch ball, and hitch pin and clip which can all be found in the etrailer Ball Mount Kit # 989900.
