The WeatherTech Cargo Liner - Cocoa # WT43356 is the correct option for your 2021 Toyota Land Cruiser when you have the 3rd row seating folded down, yes.
Please note that WeatherTech has a note in saying that their cargo mat isn't compatible with the Heritage Edition models. If that's what you have then I recommend checking out the etrailer Cargo Area Protector # e98877 as an alternative. This isn't a custom fit like the WeatherTech but it does provide a decent looking cover to help keep the back of your Land Cruiser from getting really messy. I personally used this in the back of my Chevy Equinox for a few years and have transferred it over to our Town and Country because it makes life much easier!
