There is no port to plug into on a 2009 Lexus RX 350, though if your '09 Lexus has the factory tow package you can install the 4-Pole 60" Long Vehicle End # 18002; the port you will need to tap into is in the rear cargo area of the vehicle underneath the flooring over on the driver side.

You would need to use Circuit Tester # PTW2979 to install this harness; additionally, I do strongly recommend taking a look at the linked installation video of this harness being installed on a Lexus RX 350.
