Should Transmission of 2016 Chevy Colorado be in Neutral for Flat Towing
Question:
The only thing I would like to know is after the procedure is done when towing the neutral switch is on on transfer case is the shifter also to be in neutral position when towing and not in park correct or not

I took a look at the online version of the owner's manual for your 2016 Chevrolet Colorado and on page 331 it indicates that you shift the transfer case to Neutral and then your transmission will be shifted to a few different positions before finally shifting to Park (this is for the Four-Wheel-Drive version).
You need to verify this in your copy of the owner's manual because there are a few different steps you need to take before finally shifting to Park.
Please note that a complete flat towing setup includes a base plate kit, tow bar, safety cables, vehicle tow bar wiring, a supplemental braking system, and possibly a high-low adapter. If you do not have all of these components just let me know and I can make some additional recommendations for you.
I have also attached a link to our fit guide for your Chevy Colorado which you can use to filter through all of the parts that fit your vehicle.
