According to the owner's manual for your 2002 Toyota Highlander, you will need to place the vehicle's transmission into neutral "N" and leave the key in the accessory "ACC" position when towing the vehicle with a dolly. I recommend taking a look at this for any other information you need to follow when the vehicle is in tow.

Please keep in mind that the vehicle will need to have signals coming from it even on a tow dolly unless the tow dolly itself has the signal lights. If it does not then the least invasive way to add them would be to use a set of magnetic lights for which I recommend using the Heavy Duty Magnetic Tow Lights part # TL21RK.
