The spare tire means the Malone trunk rack you asked about is not a fit for your 2012 Toyota RAV4. There is just one option for a trunk-mounted rack that will work for your vehicle, the SeaSucker Hornet Rear Window Bike Rack - Handlebar Mount - Vacuum Cup Mounted # 298-BH1302.
Apart from that, the best way to transport bikes on your 2012 RAV4 with a spare tire is a trailer hitch and hitch-mounted bike rack. For the hitch I recommend the Curt Trailer Hitch Receiver # C13406.
