Does Trailer Brake Master Cylinder Need to be Bled Before Rest of Trailer

You certainly could do that but in general it's not needed to bleed the master cylinder separately. If you wanted to though there's no reason not too. Typically it's best to just bleed the whole setup. I attached a video that shows how to bleed brakes for you to check out as well.

You can add an electric lock out to that actuator with the part # T4748800 and the brakes will stop locking up when you go to reverse the trailer. The area you have pointed out with your screwdriver cannot be used as a way to bleed the brakes though.
