Troubleshooting Single Brake Assembly of Tandem Axle Trailer not Functioning

All four wheels have their separate grounds for the electric brakes,

Since it's an electric setup and only the one brake assembly isn't working my guess is that the ground wire of that brake assembly somehow stopped making contact which is preventing just that assembly from working. Typically the assembly grounds to the trailer frame so you might check the condition of the ground and fix as needed.
