Will a 10inch drum fit behind these wheels?

A 12 inch wheel like the Kenda 5.30-12 Bias Trailer Tire with 12" Galvanized Wheel # AM30861 that you are looking at is compatible with 7 inch brake drum assemblies. A 10 inch assembly will likely be too large to fit with this 12 inch wheel.
If you want to go up to a 13 inch tire and wheel combo I recommend taking a look at the Kenda Karrier ST175/80R13 Radial Tire with 13" Galvanized Wheel # AM31994. This wheel features the same galvanized finish and 5 on 4-1/2 inch bolt pattern as the wheel you were looking at. It has a maximum load of 1,610 pounds at 65 psi.
This tire and wheel will be about 2.2 inches larger in diameter than the 5.30-12 size tire. You will just want to make sure you have the needed wheel well clearance on your trailer for the larger size. I have attached a short video demonstration on this product that you can also check out.
