Have 2015 Tundra CM, with a 1inch Lift Blocks at the Rear, will the kit work out of the box, or do I need an additional part?

Since your lift kit is a body lift there would be no need for additional spacers when installing suspension enhancement on your 2015 Toyota Tundra. The Firestone Ride Rite kit part # F2445 would fit as-is and would work well.
